Reactions 1477, p27 - 9 Nov 2013 O S Pregabalin overdose Tonic-clonic seizure: case report A 54-year-old woman developed tonic-clonic seizure after administering an overdose of pregabalin. The woman presented after ingesting 51 × pregabalin 75mg tablets (3825mg). She was alert and conscious on arrival, yet obtundation rapidly ensued with response solely to deep sternal rub. She experienced a tonic-clonic seizure at around 10 hours after overdosing; right gaze deviation and involuntary mouth movements with foaming were also noted. The self- limiting seizure persisted for around 1 minute after which obtundation returned. Lorazepam was initiated with supportive therapy; the woman regained baseline mental status, a few hours later. Her pregabalin concentration was 58 µg/mL, at around 5 hours after overdosing. Author comment: "To our knowledge, this is the first case of a seizure associated with isolated pregabalin overdose. The serum level observed in this patient is among the highest reported in the literature." Gorodetsky RM, et al. Seizures in the setting of large pregabalin overdose.
